大花蕙兰的组培苗要经过2.5～3年的营养生长，才能够进入生殖生长，进行花芽分化和发育，进而开花。进入生殖生长阶段的标志是花芽的分化。花芽的形成与叶片的数量和假鳞茎的充实度有关。足够数量的基本完成展开与伸长叶片以及充实的假鳞茎，是大花蕙兰花芽形成的必要条件（加古舜治等，1976；Vacin， 1952）。经过充分的营养生长阶段后，大花蕙兰进入生殖生长阶段。大花蕙兰的花芽由侧芽发育形成，分化为花芽的侧芽经过发育，开始抽生花葶。大花蕙兰的花芽大都形成于一年生新芽形成的假鳞茎上，这些新芽一般是上年10～11月间在老的假鳞茎上留下来的。花芽一般在假鳞茎的基部2～4节着生，每个假鳞茎上多形成1～2个花芽。
